Allopregnenolone & Increased GABA-Receptor Theory of PSSD
Hi all! I don't have time to write a whole post right now...
but, I was thinking, some of the dysphoric/anhedonic & yet Anxiolytic/numbing effects might be due to an over-active GABA-system; Post-SSRI.
So I decided to do some research...
Turns out,
SSRI's increase GABA-B receptors & binding: https://pubmed.ncbi.nlm.nih.gov/15451391/
SSRI's increase AlloPregnenolone which raises GABA: https://pubmed.ncbi.nlm.nih.gov/9807641/
GABA has negative effects/impact on sexual function via reduction of hypothalamus activity --> https://pubmed.ncbi.nlm.nih.gov/7623135/
...and Gonadotropins --> https://pubmed.ncbi.nlm.nih.gov/11394636/
and VIA the NeuroSteroids can have "paradoxical" Anhedonic (ANHEDONIA-CAUSING) and dysphoric effects --> https://pubmed.ncbi.nlm.nih.gov/21600269/
...and GABA itself has Dysphoric/Anhedonic effects --> https://pubmed.ncbi.nlm.nih.gov/33620549/

My Nitric Oxide Theory I think still trumps this but its still *interesting* either way.

And over GABA could explain memories issues... Like with Benzo or alcohol...Area1255_2021 wrote: ↑Sat Sep 11, 2021 10:28 pm Hi all! I don't have time to write a whole post right now...

Interesting.
By the way I like also the nitric oxide theory...
Re: Allopregnenolone & Increased GABA-Receptor Theory of PSSD
Yup. Just what raises allopreg? Check my theory. I hypothized this some time ago.Area1255_2021 wrote: ↑Sat Sep 11, 2021 10:28 pm Hi all! I don't have time to write a whole post right now...

Although it seems intuitive it's not ssris direct effect on 3a HSD that raises allopreg permanently in pssd
I thought we need GAMSAs like golexanolone. But thats not how to solve this.

Nope! Nitric Oxide regulates GABA, reduces Serotonin, raises Dopamine - I think that's the way to solve this!cdraham wrote: ↑Sun Sep 12, 2021 3:50 amYup. Just what raises allopreg? Check my theory. Many tried viagra or l histidine

How would you explain so many insomnias with this theory?Area1255_2021 wrote: ↑Sat Sep 11, 2021 10:28 pm but, I was thinking, some of the dysphoric/anhedonic & yet Anxiolytic/numbing effects might be due to an over-active GABA-system; Post-SSRI.
Escitalopram, 10mg/day, Jan-May 2019. Fluoxetine, May-Sept 2019. Mirtazapine 7,5mg/day, November 2019-January 2020. Escitalopram, 5mg/day, Feb-May 2020.

Because GABA inhibits serotonin levels [1].Thomas wrote: ↑Sun Sep 12, 2021 10:10 amHow would you explain so many insomnias with this theory?Area1255_2021 wrote: ↑Sat Sep 11, 2021 10:28 pm but, I was thinking, some of the dysphoric/anhedonic & yet Anxiolytic/numbing effects might be due to an over-active GABA-system; Post-SSRI.
Re: Allopregnenolone & Increased GABA-Receptor Theory of PSSD
So, this contradicts your own theory ("excessive serotonergic neurotransmission"), does it?
