desoxo-narchinol A extracted from Nardostachys jatamansi DC upregulates sert and reverses the sert inhibition of SSRIs.
"Among which, kanshone C of aristolane-type sesquiterpenoid inhibited SERT most strongly, while desoxo-nachinol A of nardosinane-type sesquiterpenoid instead enhanced SERT potently".

I'm not sure about the details like dosage or it's efficacy. All I know is I've never found a compound studied shown to reverse sert inhibition of an ssri before. I've researched pssd for years too. I think this is the key honestly I wish I could get some.
I think the effects are permanent in the sense that they upregulate the downregulated SERT in the raphe nuclei. SO theoretically you can take a round of it and upregulates your SERT and keep it upregulated as long as you don't mess them up again with SSRI.
There is no other substance that we know of that up regulates them in the Raphe. Some of the theories I've read on here speculate due to the downregulation there excess serotonin is there and has downstream effects the acts as a high serotonin feedback loop.
Cyproheptadine works to lower serotonin and upregulate 5 ht1 a receptor but it is a futile exercise without proper functioning SERT unless you take it continually which for many of us is undesirable. You can upregulate with cyproheptdatine but I think as long as your SERT in the raphe is downregulated your brain will downregulate the receptor again because of the excess.
By all means I don't think it effects everything as a cure all. I think gut microbiome and neurosteroids, epigentic expression play a role too and perhaps the treatment will have to be multi facetted to fix everything but I think if we tried this we would see improvements. I think the Sert levels in the raphe may upregulate naturally as the brain try's to achieve homeostasis but I think this push might be needed from a substance to really get to pre ssri levels.
